dc.description.abstract The main objectives of the investigation were to determine the influence of the addition of alkaline excipients to the furosemide tablet formula on the bioavailability and certain pharmacokinetic and pharmacodynamic parameters of furosemide; to determine the influence of dose reduction on the bioavailability and pharmacodynamics of furosemide, and to determine whether there was any correlation between certain pharmacokinetic and pharmacodynamic parameters of furosemide. <br><br> A literature study concerning the general characteristics of furosemide as well as the factors which influence the bioavailability , pharmacokinetics and harmacodynamics thereof, was done. As a result of the study, it was concluded that the bioavailability of furosemide could be influenced by formulation, and that it would be advantageous to ensure that the pH of the micro-environment of furosemide be made as favourable as possible during dissolution so as to improve the solubility and thus the absorption of furosemide. <br><br> A correlation between the plasma levels of furosemide, the excretion thereof in the urine and the increase in urine volume excreted within the first hour after administration was noted. Although not statistically significant, an increase in kaliuresis and natriuresis was observed in response to raised furosemide plasma levels. These observations suggest the existence of a possible correlation between certain pharmacokinetic and pharmacodynamic parameters, which might be confirmed statistically in a follow-up study with more volunteers. en
